I am trying to set up a region to place enemies like I did in VX Ace but that section doesn't seem to be there or work right.
I found the regions in the map section but it is only good for one block and doesn't seem to be doing anything.
How does this work?
I want to set up a region with enemies so the hero will encounter them when he enters that area.

I don't really understand what you're asking, but the Region settings are now under the "R" tab in the map editor (next to A, B, C, etc.) It otherwise works essentially the same way as VX Ace.

I want to have an area of about 3 by 4 squares where there are spiders attacking the group.
The R tab only lets me set up one square at a time. It also does not give any information as to what goes in that area.
Maybe I am missing something.

The R tab lets you select a region, then you can draw on the map with the pencil (MV also lets you use the circle/rectangle tool to draw, not just one tile at a time like Ace).
Then you set up the encounters on the map, changing the encounter steps if necessary, and allocate the troops to that region. -
